Colorectal cancer has long baffled scientists because, unlike most tumors, patients often do better when their cancers are packed with immune-suppressing regulatory T cells. New research finally explains why. Scientists discovered that these T cells aren’t all the same: one subtype actually helps keep tumors in check, while another shields cancer from immune attack. The balance between these “good” and “bad” cells can determine whether a tumor grows or shrinks.

Gutierrez Ruiz et al. analyzed the lysosomal proteome of pancreatic cancer cells expressing oncogenic KRAS. They identified the GEF DOCK8 as being aberrantly expressed in pancreatic cancer cells, where it controls lysosome morphology. DOCK8 regulates lysosomal actin, which impacts trafficking and secretion of lysosomal cathepsins to promote tumor cell invasion.

How cancer spreads or metastasizes is a big question for cancer researchers and patients. Mayo Clinic researchers studying pancreatic cancer—the third deadliest form of cancer in the U.S.—recently made a discovery that advances knowledge of how metastasis unfolds. They identified a cell-signaling protein that drives pancreatic cancer cell growth that could be a potential therapeutic target.

It could also help find new treatments.

The study is the first large-scale clinical trial to show that a new oral endocrine therapy outperforms the long-standing standard therapies used for early-stage HR-positive/HER2-negative breast cancer.

New research is challenging one of medicine’s oldest assumptions: that cancer must be attacked to be cured. By treating glioblastoma patients with a simple combination of resveratrol and copper, the researchers found dramatic reductions in tumor aggressiveness, cancer biomarkers, immune checkpoints, and stem-cell–related markers—all without side effects. Their approach focuses on “healing” tumors by eliminating harmful cell-free chromatin particles released from dying cancer cells, which normally inflame and worsen the disease. The findings hint at a future where inexpensive nutraceuticals could transform cancer therapy.

A Japanese-led research team has developed AUN, a groundbreaking immune-independent bacterial cancer therapy that uses two harmonized bacteria to destroy tumors even in patients with weakened immune systems. By leveraging the natural synergy between Proteus mirabilis and Rhodopseudomonas palustris, AUN selectively targets cancer cells, reshapes itself within tumors, and avoids harmful side effects like cytokine release syndrome.
